Description


As it's name suggests, worm freezing solution is added to worms before freezing them in the -80ºC incubator to ensure the survival of L1 worms.

Notes and Warning: Please note that 1M MgSO4 is added after autoclaving and the volume in the bottle will not be 1L or 2L. Be sure to add 30μL of 1M MgSO4 per 100mL in your bottle. DO NOT STORE THIS MEDIA UNTIL THE FINAL INGREDIENT HAS BEEN ADDED!

milli-Q water 710mL 1420mL
NaCl 5.8g 11.6g
1M KH2PO4 50mL 100mL
Glycerol 240mL 480mL
Add After Autoclaving
1M MgSO4 3mL 6mL

Directions

  • In a labeled beaker of the appropriate size, add a stir bar and 1/4 the final volume of milli-Q water
  • Add the appropriate volume of the reagents listed below
  • Once mixed, transfer the solution to labeled bottles
  • Autoclave the media
  • Add the remaining reagents and store at room temperature
