80% Glycerol

Description


The stock solution of Glycerol that we keep on hand is 80% (v/v) Glycerol. This is used when making a number of yeast media recipes including, YPG liquid media, YPG plates, and is used in freezing down yeast and bacterial cultures.

Media Recipe and Directions

Directions

  • You will need a labeled beaker that will fit your final volume of media and a graduated cylinder
  • Place a stirbar into the beaker and add about 50mL (500mL final volume) or 100mL (1L final volume) Milli-Q water
  • Turn on the stir bar and the hot plate (on low, the water should be warm, not hot or boiling)
  • Add the appropriate volume of Glycerol. Glycerol is incredibly viscous so be patient when pouring it out
  • When all of the glycerol is added, add milliQ water to the appropriate volume
  • Allow it to stir until incorporated. If it is not, you will see a division between water and glycerol in the beaker
  • Filter sterilize the 80% glycerol into labeled bottles
