To make dropout plates you need to measure out:
- 1.7g Yeast nitrogen Base (the one without ammonium sulfate or nutrients)
- 5g Ammonium sulfate
- 20g Sunrise (or BD) Agar
- 1.4g of the premixed SC or dropout powder
- all of which is mixed in 1L of water
- make sure that a stir bar and a little water are placed in the flask you're using beforehand so that everything will mix properly.
- after adding and mixing the ingredients, pH to 5.9
- stick the mixture in the autoclave.
- after autoclave:
add 40mL 50% glucose from our stock solution using a stipette.
