Crossing Yeast Strains
1) Clean a benchtop with 70% ethanol
2) Label appropriate number of plates as shown below:
3) Turn on flame. Using a sterile tip, gently spread the appropriate strain on the correct line. Do not allow any lines to touch.
4) Incubate plates at 30 Degrees Celsius for 2 hours (set timer)
5) After 2 hours, use sterile tip to spread the top (close) lines together (as shown in green)
6) Incubate 4 hours at 30 Degrees Celsius (timer)
7) After 4 hours, streak mixed yeast as shown above in pink
The plates are ready now for zygote picking.
8) Pick zygote and place in position as shown in purple above (dots).
Clean as you go.
