Overview

Gateway cloning is used to insert genes into a plasmid for expression. In our lab we use the Invitrogen pENTR TOPO Cloning Kit for Gateway cloning reactions.

Materials

Here you cleanly list all of the reagents and equipment in our lab or elsewhere involved in performing this protocol, with links to our wiki reagent or equipment pages if necessary (make as needed the first time you enter a protocol- a protocol is not complete without completed links to all reagents and equipment). Please include this information in a table

Procedure

This is the protocol proper; the list of step by step instructions. re-include the links to referenced equipment and reagents in-line here as they come up. If there are more than one relatively distinct sections you may break up the protocol into subsections as appropriate e.g. YEAST LYSIS and PCR. Anything which can delay the whole protocol e.g. turning on a heat block, signing up for a machine, should be listed at the top and bolded / emphasized.

Notes

Additional important or sometimes important information that is not part of the flow of steps in the protocol.

Part Numbers

list the full manufacturer name and manufacturer part number for all reagents, and then for all equipment. If these already have wiki pages you should be able to get the numbers there. PART NUMBERS will always have two sections, first Reagents and then Equipment. Please place these items in 2 tables.
