1. Find out what worm strains you need to thaw.
2. Get 1 OP-50 seeded large (10cm) NGM plate per strain to thaw worms on.
3. Label plates with the strain name, initials, date, and any other additional information about the strain.
4. Get frozen worm strains from the -80C freezer. MAKE SURE IT IS NOT THE LAST TUBE. If you are about to take the last tube, STOP and get help.
5. Use the flat end of a sterile spatula to scoop out the worm popsicle.
6. Place the plate in the incubator right side up until the liquid from the thawed worms is dry.
8. You should be able to see L1's in 48hrs, possibly 24. It is easier to see the smallest L1's by their tracks left in the bacteria than directly. If you don't see anything, keep the plates at 20C for at least a week: you may have one or two worms that will give you large numbers of F1's when they lay eggs.
