C-URA (3L) solid media! If you're making liquid, omit the agar.

0.) This can be altered for C-whatever by substituting the appropriate amino acid powder in the last ingredient of step 3. p.s. SC-ura is the same thing as C-ura, etc.

1.) Add 1.5L of water to a 4L flask and place on a magnetic stirrer / hot plate. ­­­­­­­­­­­­­­

2.) Add a stir bar whose length is about 1/3 the width of your flask bottom and turn on stirrer. No heat.

3.) Add the following ingredients to the flask:

5.1g yeast nitrogen base without amino acids and ammonium sulfate, make SURE you get yeast nitrogen base NOT yeast extract.

15g ammonium sulfate

ONLY FOR SOLID MEDIA - 60G agar (20g per liter)

4.2g C-URA amino acid powder (or whatever drop out plates you're making. You can find this on the top shelf in a basket called “DROP OUT POWDERS”)

4.) Bring the volume up to a total of 3L

5.) IF MAKING SOLID MEDIA- Adjust pH to 5.9. If you don't know how to use the PH meter please let me know.

6.) Autoclave

7.) ONLY FOR SOLID MEDIA- Add 120ml of sterile 50% glucose solution.

8.) Pour 25ml per plate with a pipet aid.
