Make a new folder with the date, who done each thing, and what the gel is.
Put the original image in the folder.
Open with GIMP
Image > Transform > Rotate 90 ° clockwise
Tools > Selection Tools > Rectangle select
Select everything except the black background.
Right click > image > crop to selection
Image > mode > greyscale
Colors > levels > auto > ok
Colors > invert
File > save as > GIMP-Edit > okay
File > export as > -EDIT > .png > hit export
At this point you should have 3 files: original, edited GIMP, edited PNG.
Right click edited png and open with inkscape. Hit ok.
Go through and make colored squares with square button on the left. Put the squares on the wells grouped per primer. Make the squares the colors that you used to label the wells.
Use the text tool on the left to put the primer number and the strain name on the boxes where the wells are.
Use the text tool to label each individual well with the MM# for the strain, WTs, and “blank”. You will want to make these small and rotate them. To rotate them click twice and click on the arrow in the corners.
Write the expected BP for the WT and deletion.
Label the hyper ladder.
Put your name, the initials of the person who ran the gel and did the PCR, and the date.
File> save as (.svg) > -labeled.
On the right click export as, change dpi settings to either 300 or 600 for higher resolution, then press export.
Print by opening the image with image viewer.
