Media Recipe and Directions

  1. Measure out 400 mg agarose in small weight boat with small scale.
  2. Pour agarose into 50 mL flask.
  3. Add Baxter water up to 20 mL line of flask.
  4. Slosh to mix agarose and water.
  5. Microwave flask without foil for 15 seconds.
  6. Swirl solution, if there is still undissolved agarose microwave in 15 second intervals until all is dissolved.

Media Recipe

For mounting worms, 20mL is sufficient. This calls for 400 mg agarose in 20 mL Baxter water.

Aliquoting and Storage Guide

You can store 1 mL agarose per flip top tube. Note: move quickly to store since this solution will solidify.

Place the tube top before rewarming and rewarm each tube at 65-75°C with 3rd spare heat block. Prep the heat block overnight if not already set.
