====== 50% Glucose ====== --- //Christine Robbins 2018/05/01 10:40// ==== Description ==== ---- The stock solution of D-glucose that we keep on hand is 50% (w/v) Glucose. This is used when making a number of yeast media recipes including, YPD liquid media, YPD plates, and all of our synthetic and drop-out media recipes. **Location:**Shelf L ===== Media Recipe and Directions===== **Directions** * You will need a labeled beaker that will fit your final volume of media and a graduated cylinder * Place a stirbar into the beaker and add about 1/4 the final volume of milliQ Water * Turn on the stir bar and the hot plate (on low, the water should be warm, not hot or boiling) * Weigh out the appropriate amount of D-glucose. You will have to use multiple weigh boats so please keep track of how much you have weighed. * SLOWLY add the glucose in. I recommend adding a little at a time and coming back when the solution is clear, then adding more. This will take a while. If you just dump in the glucose, you will create a rock that will take forever to dissolve * When all of the glucose is added, add milliQ water to the appropriate volume * Filter sterilize the 50% glucose into labeled bottles **Media Recipe** ^ Reagents ^ Final Volume |||| | ^ 500mL ^ 1L ^ 2L ^ 3L ^ | D-Glucose | 250g | 500g | 1kg | 1.5kg |
