1M Potassium Phosphate Buffer pH=6.00 is used in various medias for maintaining C. elegans including M9, NGM and SC medias.

===== Media Recipe and Directions=====

**Directions** 
  * Get a [[glassware|beaker, and bottles]] and place a stir bar in the beaker
  * Label all bottles and beaker with 1M Potassium Phosphate Buffer pH=6.00, your initials and the date
  * Add [[milliq_water|MilliQ Water]] to the beaker to about 1/4 of your final volume 
  * With the bar stiring, add the reagents
  * After all of the reagents are incorporated, [[ph_meter|pH the media]] and adjust to pH=6.00
  * Pour the media into labeled bottles (only fill to the highest marked volume on the bottle. Example: a 500mL bottle only has gradations to 400mL, so only add 400mL media) 
  * With the caps partially screwed on (you should be able to wiggle them without them coming off), add autoclave tape to he cap

  * When cool, screw caps on completely and store
